Background
SPARCL1 (secreted protein acidic and rich in cysteine-like 1) is a matricellular heparin-binding glycoprotein of the SPARC family that is secreted by quiescent endothelial cells, astrocytes and other stromal cells, where it functions as a context-dependent modulator of cell–matrix interactions, antagonizing angiogenesis by blocking endothelial proliferation and migration via its N-terminal acidic domain, stabilizing vessel integrity and limiting vascular permeability in colitis and other inflammatory states, while in the brain it promotes excitatory synaptogenesis and astrocyte differentiation, and in cancer it frequently acts as a tumor suppressor whose down-regulation accelerates invasion and metastasis yet, conversely, in gliomas or under viral injury its up-regulation can enhance inflammation by shifting macrophages toward a pro-inflammatory M1 phenotype through TLR4 activation, altogether illustrating a pleiotropic protein that integrates extracellular cues to orchestrate vascular, neuronal and immune homeostasis.
